MHC Typing of Rhesus Macaques

The last day that we will be accepting samples at the University of Wisconsin in Thursday Dec. 1, 2011. We will do our best to complete all genotyping reqeusts prior to the move as there will be downtime as the lab transfers to Miami.

Our research group has been working on MHC Typing of Rhesus macaques of Indian descent for the past several years. Nine class I alleles (in bold below) were initially chosen due to their importance in SIV epitope presentation.  Of special note, we have reported that approximately 20% of Mamu-B*17 positive and 50% of Mamu-B*08 positive macaques are able to spontaneously control replication of SIVmac239. 1,2 Two additional class II alleles, Mamu-DRB1*0306 and Mamu-DRB1*1003, have been shown to be enriched in virus infected macaque elite controllers. 3

These are the common allele names; please see Appendix A for current nomenclature and specificities of primers that include subtypes or groups.

We offer sequencing at an additional charge for confirmation of genotyping or determining specificity of group alleles. We have developed assays for additional Class II alleles, but these are all group assays that must be sequenced for allele specificity and are not offered in the standard genotyping list. Please contact the Typing Core if you are interested in a specific Class II allele .

The PCR-SSP primers we have developed are based on published sequences of Indian Rhesus MHC Class I or Class II molecules. We know that there are differences between different populations of animals. Therefore, depending on your source of animals, allele variants may be falsely detected or not show up on our tests. We cannot be responsible for these allele variants. This is the primary reason that we originally sequenced all our positive results, and that we continue to offer this confirmation at an additional charge.
